A dissolution procedure for the analysis of the metal content in Sepia officinalis melanin has been optimized and checked against a bovine liver Community Bureau of Reference (BCR) certified reference material. The content of several metal ions has been determined by GF-AAS and ICP-AES techniques in samples of Sepia officinalis melanin and the results have been compared with those obtained after a harsh treatment of the melanin with 6 M HCl. Remarkable differences not only on metal distribution but also structural changes induced in the melanin polymer by the acidic treatment have been pointed out and underlined by C-13 CPMAS NMR spectroscopy.
