Introduction: Bud-derivatives, that represent a category of natural products obtained macerating meristematic tissues of trees and plants, are very expensive products compared to other botanicals, since the collection period of their raw materials is extremely limited over time1. In this study, a re-use procedure based on an innovative green extraction to valorize Castanea s. bud by-products is presented. Methods: Pulsed Ultrasound-Assisted Extraction2 (PUAE) has been employed to extract further valuable material from bud by-products. UV-Vis spectrometry coupled with chemometrics were employed to screen the best experimental conditions. Phytochemical fingerprints by HPLC have been used to investigate the most promising extracts comparing them with the corresponding commercial Glyceric Macerate (GM). Results: Castanea s. wet marcs, remaining after the production of the corresponding GM, were extracted using their same solvent by PUAE. PUAE process conditions have been optimized by Design of Experiment (DOE) using a 24-1 fractional factorial design (duty cycle 80%, extraction time 15 minutes and solvent/ratio 1/10). As response variable of the DOE, the score on the first principal component calculated by the PCA (Principal Component Analysis) performed on the UV-Vis spectra, was taken into account (untargeted fingerprints). A targeted phytochemical fingerprint by HPLC3 of the most promising extract selected by the DOE was performed. This extract, whose chromatographic profile is very similar to the corresponding commercial GM, has a content in secondary metabolites of 160.42 mg/g of fresh weight marcs, which represent about the 12% of the commercial GM (1276.2 mg/g of fresh weight buds). Conclusions: This study takes place in the context of an Alcotra Italy-France trans-frontier project called FINNOVER (2017-2020)4. The results obtained show an innovative extraction and re-use strategy, applied to Castanea s. bud production as case study, to obtain value-added products from food supplements by-products in alternative to incineration or composting.
