The possibility of developing a system for the cultivar genetic certification of grapevine and chestnut is discussed and a method of multiplex analysis is tested and proposed in order to reduce the cost of the analysis. Microsatellite (=Simple Sequence Repeats) markers, being considered particularly suitable for the DNA typing of cultivars of fruit species and grapevine (although unable to identify varieties originated by somatic mutation), were isolated in chestnut and the primers designed already taking into account the characteristics required for the use of the loci in multiplex analysis. This technique is based on the analysis at once of several (6-13) loci mixed together, using a semi-automated DNA sequencer and the fluorochrome technology for the primer labeling. Multiplex PCR was set up for grapevine microsatellite published loci which were successfully amplified and analysed in groups of 5.
