Cell nuclei were isolated from leaf tissue of different species of Capsicum and the relative fluorescence intensity was measured by flow cytometry after propidium iodide staining. Pea cells with known nuclear genome size (7.09 pg/nucleus) were used as standard to determine the nuclear content of the samples. The found DNA content ranged between 7.65 (C. annuum) and 9.72 pg (C. pubescens), being the general mean of the genus 8.42 pg. According to their DNA content, it was possible to divide the studied species into three main groups. The contribute of the results to a better understanding of systematic relationship within the genus Capsicum is briefly discussed.
