NG2-expressingparenchymalprecursors(NG2+p)serveasprimarysourceofmyelinatingoligodendrocytesinboththedevelopingandadultCentralNervousSystem(CNS).However,theirabundance,limiteddifferentiationpotentialatadultstagesalongwithstereotypicreactiontoinjuryindependentoftheextentofmyelinlosssuggestthatNG+pexertfunctionsadditionaltomyelinproduction.Insupportofthisview,NG2+pexpressacomplexbatteryofmoleculesknowntoexertneuromodulatoryandneuroprotectivefunctions.Further,theyestablishintimatephysicalassociationswiththeotherCNScelltypes,receivefunctionalsynapticcontactsandpossessionchannelsapttoconstantlysensetheelectricalactivityofsurroundingneurons.TheselatterfeaturescouldendowNG2+pwiththecapabilitytoaffectneuronalfunctionswithpotentialhomeostaticoutcomes.HerewesummarizeanddiscusscurrentevidencefavoringtheviewthatNG2+pcanparticipateincircuitformation,modulateneuronalactivityandsurvivalinthehealthyandinjuredCNS,andproposeperspectivesforstudiesthatmaycompleteourunderstandingofNG2+prolesinphysiologyandpathology.
