An in vitro test of cell-mediated cytotoxicity (CMC) in the rat is described. Yarget cells were peritoneal macrophages, labeled vith 51Cr, adhering to the plastic surfacce of microtiter plates. The spontaneous 51Cr release was between 1.5 and 2.5% of maximum release/hr. Kinetics, immunological specificity of target cell lysis, and the effect on CMC of various anti-target cell antisera show that the target peritoneal macrophage is comparable to other types of well established target cells for study of the CMC in vitro.
