The aim of this work was to test if the dilution of thawed dog semen with prostatic fluid (PF) can revert capacitation and then increase semen longevity. One ejaculate was collected from each of 15 adult healthy dogs, raw semen was immediately assessed for motility and concentration and then centrifuged. Seminal plasma was collected into a sterile vial and stored frozen at -25°C. Diluted sperms was frozen over nitrogen vapours.Thawed semen was diluted 1:2 with either a TRIS buffer or with the corresponding PF, both pre-warmed at 38°C, then subjected to analysis. The possible revertion of capacitation of spermatozoa is not confirmed by the results relative to acrosome status, which were not different between the two dilution treatments, both at hr 0 and 2 post-thawing. Besides, PF does not prolong post-thaw semen longevity since motility was significantly higher in PF-diluted samples at hr 0 but lower during the following hrs and decreased to less than 5% at hr 4 post thawing.
