Production of H2O2 was stimulated in cell-free crude homogenates from callus of melon, treated with hyphal wall components of Fusarium oxysporum f.sp. melonis. Active oxygen production was detected as PO-catalized oxidative bleaching of a fluorescent probe (pyranine) occurred when the homogenate of a resistant melon genotype was treated with HWC of an avirulent race of Fusarium, but not when a non resistant plant genotype or a virulent race of the fungus were included in the incubation mixture. The role of a NADPH-dependent membrane oxidase and the role of a LOX/fatty acid system were discussed.
